Electrophoretic Study on Seed Storage Proteins of Groundnut (Arachis hypogaea L.) Cultivars of India

Abstract

Electrophoretic separation of seed storage protein of 70 Indian released groundnut cultivars was studied on SDSPAGE
to find distinction in banding pattern and to explore the possibility to use the traits to meet DUS (Distinction,
Uniformity and Stability) testing. Buffer soluble proteins from seeds were extracted using Tris-HeI buffer and
subjected to SDS-PAGE. Thirty-six distinct bands with molecular weight ranging from 7 to 128 kD were resolved
and polymorphism was observed for the banding pattern. The cluster analysis based on cosine coefficient ofassociation
showed distinction of individual cultivars, indicating that seed storage protein trait can be supplemented to the
morphological traits for DUS testing in groundnut.
